Περίληψη: Postmenopausal osteoporosis is a common metabolic bone disorder characterized by estrogen deficiency, increased bone turnover, and progressive bone loss.sesamol is now described as a natural phenolic lignan possessing well-documented antioxidant and anti-inflammatory properties, while glyburide is introduced as a sulfonylurea antidiabetic agent with reported anti-inflammatory and metabolic regulatory effects. The present study evaluated the osteoprotective effects of sesamol and glyburide, administered individually and in combination, in an ovariectomized (OVX) rat model of postmenopausal osteoporosis. Adult female Sprague–Dawley rats were randomly assigned to eight experimental groups: normal control, OVX control, sesamol low-dose, sesamol high-dose, glyburide low-dose, glyburide high-dose, combination treatment (sesamol + glyburide), and alendronate-treated groups. Following 30 days of treatment, bone-related parameters were evaluated by histopathological examination of the femur, determination of apparent bone mass density and femoral ash content, and serum biochemical analyses, including alkaline phosphatase (ALP), calcium, phosphorus, and creatinine. Compared with the normal control group, ovariectomized rats exhibited deterioration of bone microarchitecture, reduced apparent bone density and femoral ash content, and altered serum biochemical parameters. Treatment with sesamol, glyburide, and their combination significantly improved these parameters compared with the OVX control group. Histopathological examination also demonstrated qualitative improvements in cortical and trabecular bone architecture in the treated groups. Alendronate produced the greatest improvement among the treatment groups. In conclusion, sesamol and glyburide attenuated ovariectomy-induced alterations in bone structure and serum biochemical parameters in this experimental model. These findings provide preliminary preclinical evidence supporting further mechanistic investigations and long-term preclinical studies to evaluate the therapeutic potential of these interventions in postmenopausal osteoporosis.
