| Περίληψη: | Frequent fires threaten amphibian population persistence worldwide. Conservation translocation after large wildfires and natural exposure during dispersal after small fires represent two common post-fire recovery scenarios in amphibians. However, skin microbiota responses and recovery trajectories following different ash exposures remain poorly understood. This study used semi-natural enclosures to simulate these two post-fire recovery scenarios, assessed hepatic oxidative stress and inflammation-related gene expression at week 7, and used 16S rRNA gene sequencing to track the 7-week recovery dynamics of Rana dybowskii skin microbiota. The results showed that hepatic oxidative stress biomarkers did not differ among groups at week 7. Skin microbiota analyses revealed significant ash source dependence and the interaction between ash source and recovery time during recovery. In the conservation translocation experiment, the wildfire ash-disturbed microbiota showed high resilience, shifting from deterministic to drift-dominated assembly and converging toward the time-matched control trajectory by week 7. In the comparative ash exposure experiment, straw ash caused persistent diversity loss and structural divergence over 7 weeks, indicating a low-diversity divergent trajectory associated with deterministic selection and priority effects. These findings suggest that timely translocation to clean habitats may facilitate skin microbiota recovery after wildfire ash exposure in R. dybowskii. In contrast, straw ash was associated with persistent microbiota divergence and a path-dependent recovery lag during the 7-week observation period. This study reveals post-fire recovery dynamics of amphibian skin microbiota across exposure scenarios and highlights the need to integrate microbial endpoints into post-fire amphibian conservation and ash-related risk assessment.
