Περίληψη: The co-occurring type 2 diabetes mellitus and dyslipidemia is a thermal treatment problem due to the interrelated pathophysiology of insulin resistance, hyperglycaemia, and lipid metabolic dysregulation. The traditional use of monotherapy or independent use of antidiabetic and lipid lowering drugs often leads to poor solubility, reduced oral bioavailability, pharmacokinetic discrepancy and reduced compliance by the patient. In order to overcome these shortcomings, the current research focuses on a liquid antisolvent precipitation (LASP)-designed dual nanoparticle system that is designed to co-deliver glibenclamide and atorvastatin, two BCS Class II compounds with an inherently poor aqueous solubility. Same Preformulation testing such as fourier -transform infrared spectroscopy (FTIR), solubility profiling, differential scanning calorimetry (DSC) identified the identity of drugs and excipients compatibility. The LASP method was used to generate nucleation by using rapid supersaturation, thereby producing nanosized particles, which were stabilized using Poloxamer 188 to avoid aggregation. The main method of systematic optimisation was done through the manipulation of solvent-antisolvent ratios, stirring intensity, and stabiliser concentration. Thorough characterisation, which included particle size, polydispersity index, zeta potential, morphological examination and crystallinity studies indicated successful characterisation of nanocrystals with favourable physicochemical characteristics. In-vitro solubility and dissolution analysis have shown a significant difference in terms of dissolution rate in comparison to the raw drug powders. Both physical and chemical stability of the formulation was confirmed using stability tests under International Conference on Harmonisation (ICH) conditions. The dual nanoparticle platform is an intellectually justified method of simultaneous regulation of glycaemic and lipid regulation with the ability to minimise polypharmacy, enhance bioavailability, and enjoy simultaneous pharmacokinetic behaviour. This strategy has potential to provide hope in the future as a way of translating into future therapies on metabolic disorders; it is a promising approach to strike on the multifactorial nature of the issue of diabetes-related dyslipidaemia.
