The culturability was studied by means of standard plate count method so the E. coli cells were cultured in Yeast and Chromocult Coliform Agar and the E. faecalis cells in Brain Heart Infusion and Chromocult Enterococcus Agar. The viability, direct viable count procedure, consisted of exposing bacterial cells to a combination of SYTO 9 green fluorescent nucleic acid stain and propidium iodide (PI) red fluorescent nucleic acid stain, which were used to differentiate the alive from the dead cells. The FISH method was preformed usingthe EUB338I probe specific for E. coli 683 CECT and E. faecalis 481 CECT. To monitor the survival of E. coli and E. faecalis in FISH and viability methods an epifluorescence microscopy was used. Accuracy and detection limit of the epifluorescence microscopic viability- FISH procedure were investigated. The method was then applied to sea water samples. The culturability and viability methods which were applied to the sea water samples are not significant as the correlation coefficient is 0.03 (P > 0.05), fact that implies that the cells through the days lost the culturable state ( 98% for E. coli and 95% for E.faecalis) but they could store the membrane integrity as it comes from the results of viability.
