Description: ENG- Pig production is a key sector of the agri-food economy in Spain and the European Union. Reproductive efficiency is essential to ensure its competitiveness, and artificial insemination is currently the main reproductive tool used in pig production. The success of this process largely depends on semen quality, which must be preserved and transported under optimal conditions. Unlike other species, boar sperm do not tolerate freezing well. For this reason, semen is usually stored at 17 °C. This system allows good sperm quality to be maintained, but only for a limited period, as quality progressively declines over time. Understanding why sperm lose viability over the days is essential to improve preservation protocols, extend the shelf life of semen doses, and increase productivity in the pig sector. This Doctoral Thesis analyses the molecular mechanisms that regulate the ability of pig sperm to withstand storage at 17 °C. One of the key factors is calcium, an essential molecule for sperm function but potentially harmful if not properly regulated. The results show that an excess of intracellular calcium negatively affects sperm quality, whereas adequate regulation of intracellular calcium alone is sufficient to maintain sperm functionality during storage. The Thesis also examines the role of other relevant cellular mechanisms. On the one hand, the protein HSP70 contributes to maintaining a low metabolic profile of sperm during storage, which is crucial for preserving viability. On the other hand, the activation of caspases—proteins involved in programmed cell death—was found to limit sperm survival. Inhibition of these proteins improves the motility and viability of stored semen. Overall, this Thesis demonstrates that the preservation of boar semen depends on a precise balance between calcium regulation, metabolic control, and the inhibition of cell death processes. These findings provide fundamental knowledge and open the door to new strategies to improve semen preservation, with a direct impact on productivity and efficiency in the pig production sector
