Περίληψη: Mercury (Hg) is a toxic metal commonly measured in whole blood. Hg toxicity is dependent on the chemical species, where exposure to methylmercury (MeHg) tends to be more harmful than inorganic Hg (iHg), i.e., Hg2+ or Hg0. Nonetheless, the analysis for both total Hg and Hg speciation in blood are important for environmental and clinical investigations. This work focuses on developing a validated, automated, fast, and reliable method for both total Hg and the separation of iHg and MeHg in whole blood within the same analytical system. The optional addition of vapor generation (VG) was investigated to explore the potential for lower detection limits with a total of four analysis modes in one automated system, i.e., total Hg without VG, total Hg with VG, Hg speciation without VG, and Hg speciation with VG. The method presented here uses a C18 column for the separation of the Hg species within ∼7 minutes. Each of the four analysis modes were evaluated for accuracy by analyzing NIST SRM 955c and 955d, and archived blood samples distributed in the New York State Department of Health (NYS DOH) Biomonitoring Proficiency Testing program for trace elements. While VG did not result in a lower method limit of detection (LOD), performance was deemed reasonable. The method LODs without VG in a low-level blood reference material were 0.8 µg L−1 and 0.7 µg L−1 for iHg and MeHg, respectively, compared to 1.3 µg L−1 and 0.8 µg L−1 with VG. [ABSTRACT FROM AUTHOR]
