| Περίληψη: | Vanadium is widely detected in groundwater and poses significant risks to human health at elevated concentrations. Microbial remediation offers a promising and effective approach for vanadium treatment. However, vanadate [V(V)]-reducing microorganisms have mainly been characterized using pure metal-reducing strains, leaving the tolerance and reduction potential of indigenous groundwater microorganisms unclear. In this study, an efficient V(V)-reducing bacterium was isolated from vanadium-contaminated groundwater using an enrichment culture method. The isolated strain was then identified as Lysinibacillus fusiformis CUGB1 by combining 16S rDNA phylogenetic analysis with its biological features. Whole-gene sequencing was further performed to characterize the function of this strain. Up to 89.6% V(V) was removed in 120 h, producing the amorphous tetravalent vanadium precipitates. Microbial transcriptomic analysis under 10 mg/L V(V) revealed that functional genes related to carbohydrate synthesis, peptidoglycan biosynthesis, and spore coat formation (e.g., rpmD, cotD, cotY_Z) were upregulated under vanadium stress, suggesting a strategy of sporulation to resist vanadium. The elevated levels of membrane interfacial electron transfer compounds (e.g., cytochrome c, nicotinamide adenine dinucleotide, glutathione, extracellular polymeric substances) along with transmembrane electron shuttles (e.g., riboflavin and quinones) promoted electron transfer, thereby supporting V(V) reduction. This species was widely distributed in natural environments on Earth, displaying great bioremediation potential. This study expanded the family of known V(V) reducing microorganisms and contributed to the sustainable bioremediation of vanadium-contaminated groundwater. (Copyright © 2026 Elsevier B.V.
